Phenylketonuria (PKU) is a rare autosomal recessive metabolic disorder characterized by a mutation in the gene encoding the enzyme phenylalanine hydroxylase (PAH). This deficiency prevents the conversion of the essential amino acid phenylalanine (Phe) into tyrosine, leading to toxic accumulations of Phe in the blood and brain. Untreated PKU results in severe intellectual disability, cognitive deficits, and neurological complications.
Because phenylalanine is present in almost all natural dietary proteins, PKU management requires a strict, lifelong restriction of dietary protein intake, supplemented by Phe-free amino acid medical foods. Traditional free amino acid mixtures often suffer from rapid absorption rates compared to intact proteins, resulting in a spike in blood amino acid levels and subsequent oxidation of the amino acids rather than utilization for protein synthesis. Additionally, the highly bitter taste and strong odor of free amino acids lead to low compliance among pediatric and adolescent patients.
To solve these challenges, SINOFN's R&D division has developed specialized delivery matrices that mask taste and control absorption kinetics. By utilizing advanced coating technologies and investigating peptide-bound configurations, we ensure that our PKU formulations support stable plasma amino acid levels, promote optimal nitrogen balance, and improve patient compliance across all age groups.
A key focus of SINOFN's technology roadmap is the transition from free amino acids to peptide-bound delivery formats.
